The dynamic properties of unit responses to amplitude-modulated tones were studied using modulation with pseudorandom noise and described by cross-covariance and integrated cross-covariance functions between the discharge rate and the modulation. Under the experimental conditions used, these two functions are valid approximations of the system's impulse and step response functions respectively. 2. On the basis of their impulse response functions units could be classified into two groups, Type I with a low adaptation and Type II with a large degree of adaptation as well as a damped oscillation in their impulse response functions. 3. The response pattern of the Type II units is most likely the result of a negative feed-back striving to keep the discharge rate at a nearly constant level. 4. The cross-covariance functions are shown to remain unchanged during long duration recordings from the same unit. << Less

Using cDNA of chick chondroitin 6-sulfotransferase (C6ST), human C6ST cDNA has been isolated. The amino acid sequence of human C6ST displayed 74% identity to chick C6ST. The major difference in amino acid sequence between chick C6ST and human C6ST was the presence of a unique hydrophilic domain in human C6ST. A 7.8-kb message of C6ST was expressed ubiquitously in various human adult tissues, indicating a rather diverse function of C6ST. << Less

Chondroitin 6-sulfotransferase (C6ST) catalyzes the transfer of sulfate to position 6 of the N-acetylgalactosamine residue of chondroitin. To obtain direct evidence regarding the function of C6ST and its product, chondroitin 6-sulfate, in vivo, we isolated the mouse C6ST gene (C6st) and generated mice deficient in this gene (C6st(-/-)) by embryonic stem cell technology. C6st(-/-) mice were born at approximately the expected frequency and were viable through adulthood. In the spleen of C6st(-/-) mice, the level of chondroitin 6-sulfate became almost undetectable. Analyses of these knockout mice provided insights into the biosynthesis of oversulfated chondroitin sulfates in mice; chondroitin sulfate D in the brain of null mice and the cartilage and telencephalon of null embryos disappeared, whereas the chondroitin sulfate E level in the spleen and brain of the null mice was unchanged. Despite the disappearance of chondroitin sulfate D structure, brain development was normal in the C6st(-/-) mice. Further analysis revealed that the number of CD62L(+)CD44(low) T lymphocytes corresponding to naive T lymphocytes in the spleen of 5-6-week-old C6st(-/-) mice was significantly decreased, whereas those in other secondary lymphoid organs were unchanged. This finding suggested that chondroitin 6-sulfate plays a role in the maintenance of naive T lymphocytes in the spleen of young mice. << Less

Chondroitin 6-sulfotransferase (C6ST) catalyzes the transfer of sulfate from 3'-phosphoadenosine 5'-phosphosulfate to position 6 of the N-acetylgalactosamine residue of chondroitin. Using chick C6ST cDNA as a probe, we cloned the cDNA of mouse C6ST. The mouse enzyme was predicted to be composed of 472 amino acids, and exhibited 71% sequence identity with the chicken enzyme. The mouse and chicken catalytic domains exposed to the luminal side exhibited 81% identity, while the homology of the remaining regions was less. Transfection and expression of the mouse cDNA in COS-7 cells yielded C6ST activity. Keratan sulfate sulfotransferase activity, which was simultaneously expressed, amounted to 3% of the C6ST activity, this value being significantly lower than that observed in the case of the chicken enzyme. Mouse C6ST mRNA was strongly expressed in the spleen, lung, and eye. In situ hybridization revealed that the transcript was localized in stromal cells in the marginal zone and red pulp of the spleen, and stromal cells in the bone marrow. Fluorescence in situ hybridization analysis revealed the gene is located in mouse chromosome 9. << Less
